It is a well-established fact that as we grow older, our bones become more brittle and prone to fracturing. It is also well established that loss of mass is a major reason for older bones fracturing more readily than younger bones, hence medical treatments have focused on slowing down this loss. However, new research from scientists at the U.S. Department of Energy (DOE)'s Lawrence Berkeley National Laboratory (Berkeley Lab) shows that at microscopic dimensions, the age-related loss of bone quality can be every bit as important as the loss of quantity in the susceptibility of bone to fracturing.
Using a combination of x-ray and electron based analytical techniques as well as macroscopic fracture testing, the researchers showed that the advancement of age ushers in a degradation of the mechanical properties of human cortical bone over a range of different size scales. As a result, the bone's ability to resist fracture becomes increasingly compromised. This age-related loss of bone quality is independent of age-related bone mass loss.
"In characterizing age-related structural changes in human cortical bone at the micrometer and sub micrometer scales, we found that these changes degrade both the intrinsic and extrinsic toughness of bone," says Berkeley Lab materials scientist Robert Ritchie. "Based on multiscale structural and mechanical tests, we attribute this degradation to a hierarchical series of coupled mechanisms that start at the molecular level."
Ritchie, who holds joint appointments with Berkeley Lab's Materials Sciences Division and the University of California (UC) Berkeley's Materials Science and Engineering Department, is the senior author of a paper published in the Proceedings of the National Academy of Science (PNAS) that describes this work. The paper is titled "Age-related changes in the plasticity and toughness of human cortical bone at multiple length scales."
Co-authoring the PNAS paper with Ritchie were Elizabeth Zimmermann, Eric Schaible, Hrishikesh Bale, Holly Barth, Simon Tang, Peter Reichert, Bj?rn Busse, Tamara Alliston and Joel Ager.
Human cortical or compact bone is a composite of collagen molecules and nanocrystals of a mineralized form of calcium called hydroxyapatite (HA). Mechanical properties of stiffness, strength and toughness arise from both the characteristic structure at the nanoscale, and at multiple length scales through the hierarchical architecture of the bone. These length scales extend from the molecular level to the osteonal structures at near-millimeter levels. An osteon is the basic structural unit of compact bone, comprised of a central canal surrounded by concentric rings of lamellae plates, through which bone remodels.
"Mechanisms that strengthen and toughen bone can be identified at most of these structural length scales and can be usefully classified, as in many materials, in terms of intrinsic toughening mechanisms at small length scales, promoting non-brittle behavior, and extrinsic toughening mechanisms at larger length scales acting to limit the growth of cracks," Ritchie says. "These features are present in healthy, young human bone and are responsible for its unique mechanical properties. However, with biological aging, the ability of these mechanisms to resist fracture deteriorates leading to a reduction in bone strength and fracture toughness."
Working with the exceptionally bright beams of x-rays at Berkeley Lab's Advanced Light Source (ALS), Ritchie and his colleagues analyzed bone samples that ranged in age between 34 and 99 years. In situ small-angle x-ray scattering and wide-angle x-ray diffraction were used to characterize the mechanical response of the collagen and mineral at the sub micrometer level. A combination of x-ray computed tomography and in situ fracture-toughness measurements with a scanning electron microscope were used to characterize effects at micrometer levels.
"We found that biological aging increases non-enzymatic cross-linking between the collagen molecules, which suppresses plasticity at nanoscale dimensions, meaning that collagen fibrils can no longer slide with respect to one another as a way to absorb energy from an impact," Ritchie says. "We also found that biological aging increases osteonal density, which limits the potency of crack-bridging mechanisms at micrometer scales."
These two mechanisms that act to reduce bone toughness are coupled, Ritchie says, in that the increased stiffness of the cross-linked collagen requires energy to be absorbed by "plastic" deformation at higher structural levels, which occurs by the process of micro cracking.
"With age, remodeling of the bone can lead the osteons to triple in number, which means the channels become more closely packed and less effective at deflecting the growth of cracks," he says. "This growing ineffectiveness must be accommodated at higher structural levels by increased micro cracking. In turn, the increased micro cracking compromises the formation of crack bridges, which provide one of the main sources of extrinsic toughening in bone at length scales in the range of tens to hundreds of micrometers. Thus, age-related changes occur across many levels of the structure to increase the risk of fracture with age."

NEW YORK (AP) ? When third-seeded Maria Sharapova looks at the list of top contenders for the U.S. Open, here's what she sees:
?Serena Williams, seeded 28th and on the comeback trail, and her sister, Venus, unseeded and barely on the radar this year.

Clijsters' withdrawal was the latest bit of news that ? on paper, at least ? appeared to open things up for Sharapova, a three-time major winner who, of late, has been playing her best tennis since shoulder surgery derailed her in late 2008.
"I can't really think like that," Sharapova said when asked how Clijsters' absence might help her chances for a second title at Flushing Meadows. "I don't think that's a mindset of a winner, to be honest. You've got to be ready to face anyone at any given moment."
On Monday, Sharapova will open against Heather Watson, the 104th-ranked 19-year-old from Britain making her U.S. Open debut.
Also opening play Monday is fifth-seeded Petra Kvitova, who beat Sharapova in the Wimbledon finals earlier this summer, further cementing the argument that there are no sure things, or dominant players, at the current time in women's tennis.
"She was able to find an answer, you know, in things that I kind of challenged her with," Sharapova said. "It was a really great match for her at a big stage. That's the only way you can really look at it."
And yet, since that 6-3, 6-4 win over Sharapova at Wimbledon, Kvitova has won a total of two matches.
"I think she's a good enough player to find her form back here," Sharapova said.
After missing the better part of a year with a series of ailments that started when she got cut by glass at a restaurant in July 2010, Serena Williams is rounding into form. Earlier this month, she won tournaments in Stanford and Toronto and is 16-2 since June. Even as the 28th seed, she is widely considered the woman to beat.
"I'm just here to play one match, and the next match, and hopefully I can get to seven wins," Williams said last week.
Play was set to begin largely on schedule Monday despite Hurricane Irene, which washed out practice days over the weekend. The only exception is in Arthur Ashe Stadium, where the first match will begin at 1 p.m. instead of 11 a.m., as workers prepare the stadium, which had been battened down for the hurricane.

BEIJING (Reuters) ? China's state news agency on Thursday condemned a Pentagon report on China's growing military might, calling its account of Beijing's weapons modernization drive an alarmist "cock-and-bull story."
The Pentagon's annual assessment of China's military strength, which was released on Wednesday, said Beijing appeared to be on track to forge a modern military by 2020, a build-up that could destabilize the Asia-Pacific region.
The Chinese government has yet to give its official response, which is likely to be tersely dismissive, but the state-run Xinhua news agency made a starting shot.
"The allegation is an utterly cock-and-bull story about the Chinese military based on a wild guess and illogical reasoning," an English-language commentary published by Xinhua said.
"China, which has adhered to a defensive military policy, with its rising economic clout and sprawling commercial and strategic interests around the world, has every right to build a competent military," the commentary said.
The Pentagon's report was released days after U.S. Vice President Joe Biden finished a visit to China dominated by mutual vows of cooperation and goodwill. The annual quarrel over the Pentagon report could, at least briefly, dampen the upbeat tone set by Biden's visit, which focused on economic issues.
Fueled by China's booming economy, the People's Liberation Army's weapons upgrades in the past decade have exceeded many earlier U.S. forecasts. China's aircraft carrier program, cyber warfare capabilities and anti-satellite missiles have unsettled neighbors and Washington.
But China sees its military modernization as a rightful extension of its growing economic status, and it staunchly defends is military spending as defensive in nature.
The Pentagon flagged concerns about Beijing's widening military edge over Taiwan, the self-ruled island that China calls an illegitimate breakaway.
Despite "large quantities of antiquated hardware and a lack of operational experience," the U.S. report said the People's Liberation Army is closing the technological gap with modern military forces.
Xinhua called the Pentagon's conclusions "much ado about nothing," and said Chinese people thought it "baffling" that the U.S. could criticize China when its own military spending was 40 percent of the world's total in 2010.
China put its defense budget for 2011 at 601.1 billion yuan ($91.5 billion), up from 532.1 billion yuan last year, or a 12.7 percent increase. But many experts believe China's actual spending on the 2.3 million-strong PLA is far higher than what the government reports.
By comparison, the Pentagon rolled out a record base budget for fiscal year 2012 of $553 billion, up $22 billion from the level enacted for 2010.
Xinhua cited improving military ties between China and the U.S., a development it said should be embraced.
"The two countries should cherish their hard won improved bilateral ties, particularly the military relations, instead of blaming and smearing each other," the news agency said.

This is no bad thing as the original played so well - and most fans of the sport will just be happy with the welcome ability to play a rugby game one vs one online for the
first time.
Like buses, you wait for one rugby game and two come at once with the promising looking Jonah Lomu's Rugby Challenge due next month.
But for the meantime at least, this does a great job of filling a gaping gap for sports gaming fans.

Last week, a frequent contributor to a jihadist website posted the threat against Letterman. He urged Muslim followers to "cut the tongue" of the late-night host because of a joke and gesture the comic had made about al-Qaida leaders on his CBS show earlier this summer.
"A guy, a radical extremist threatened to cut my tongue out," Letterman marveled during Monday's monologue. Then, referring to his disastrous turn hosting the 1995 Oscarcast, he added, "I wish I had a nickel for every time a guy has threatened (that). I think the first time was during the Academy Awards."
"And so now," he continued, "State Department authorities are looking into this." But they could save themselves some time: "Everybody knows it's (Jay) Leno."
Along with his monologue, Letterman mined the situation for his Top Ten List: "Top Ten Thoughts That Went Through My Mind After Hearing about the Threat."
One joke that may have helped spark the fatwa was one of several lampooning al-Qaida in Letterman's June 8 monologue. This was just days after the death of al-Qaida leader Ilyas Kashmiri, who was killed by a U.S. drone strike in Pakistan. Though Kashmiri was rumored to be a long-shot choice to succeed Osama bin Laden, he wouldn't have worked out even had he lived, Letterman cracked, pointing to Kashmiri's "rocky start" as a front-runner: "He botched up the story of Paul Revere."
The real butt of that joke: Sarah Palin, potential 2012 GOP presidential candidate, who in early June on her "One Nation" bus tour had claimed that Paul Revere's famous ride was intended to warn British soldiers as well as his fellow colonists.

The private firm, part of the Site Intelligence Group, provides information to government and commercial clients on what jihadists are saying on the Internet and in traditional media. Raisman said the online forum is often used by al-Qaida.
The FBI said last week that it was looking into the threat.
While Letterman and his writers were polishing their jokes Monday afternoon, outside, a bomb-sniffing dog was led around the periphery of the Ed Sullivan Theater in midtown Manhattan. Meanwhile, ticket-holders queuing up along Broadway seemed relaxed about attending Letterman's first taping since the assassination threat. Some were even unaware that his life had been threatened.

NEW YORK (Reuters Health) ? A new study adds to evidence that bone drugs may play a role in suppressing cancer development, although there is still no proof that taking the medications would stave off malignancies.
The drugs, called bisphosphonates, have generated both good and bad headlines recently. Some work has linked them to fewer cases of cancer, while other research has found an increased risk of certain thigh fractures and serious jaw problems among women who take the drugs to treat the bone-thinning disease osteoporosis.
Bone drugs on the market include Merck's Fosamax, Roche's Boniva, Novartis's Reclast and Warner Chilcott's Actonel.
In the new study, researchers found that women on Actonel were only half as likely to be diagnosed with colon cancer as those who'd gotten just one prescription for the drug or none at all.
"At this point, I think people who are already taking the drug can be reassured," Dr. Harminder Singh, who worked on the study, told Reuters Health.
But he added that it's too soon to think about taking Actonel to prevent cancer.
"For people to start taking it, there are a few other things that need to be worked out -- the cost and the side effects," said Singh, of the University of Manitoba in Canada.
According to the American Cancer Society, one in 19 men develops colorectal cancer at some point, and slightly fewer women do. The disease is the third leading cause of cancer deaths in the U.S.
An Israeli study published earlier this year found a link between bisphosphonates and a lower risk of colon cancer among women, but it wasn't clear how to explain that result. It could be that the drugs have protective effects, or it could be that women taking them are healthier overall.
In contrast to the Israeli work, Singh's study, which is published in the journal Cancer, takes into account how often people see their doctors -- one factor that might muddy any differences found between those who take bone drugs and those who don't.
The research team tapped into data from a cancer registry in Manitoba, comparing colon cancer patients to people without the disease, but of the same age and sex.
About 1.1 percent (264) of the nearly 25,000 women in the comparison group had been given Actonel by their doctor, while only 0.6 percent (15) of 2,493 women with colon cancer had gotten the drug.
One way to interpret those findings is to chalk the difference up to a protective effect of Actonel. Indeed, women on that medication had 45 percent lower odds of having colon cancer than those who didn't take it.
But with a study design where you only observe patients, you can't ever be certain why some fare better than others, Singh cautioned.
"The ultimate proof of these agents would be a randomized controlled trial," he explained.
What's more, the researchers didn't have enough data to see if the findings would hold up for men, and they didn't find any reliable effects for the other bone drugs.
Even if Actonel did afford some protection against tumors, it's not cheap and its long-term side effects are not well known. The U.S. Food and Drug Administration is currently reviewing evidence suggesting bone drugs might raise the risk of esophageal cancer.
A month's supply of Actonel generally costs over $100.
So far, quitting smoking is one of the only things that experts agree will lower the risk of cancer. For colon cancer in particular, the U.S. Preventive Services Task Force, a federally supported expert panel, advises screening all people between 50 and 75 using stool tests, sigmoidoscopy or colonoscopy.
